ABSTRACT

Being vector of West Nile Virus and falariasis the control of Culex quinquefasciatus is likely to be essential. Synthetic insecticide treatment is looking most effective for vectors mosquito control. However, these products are toxic to the environment and non-target organisms. Consequently, ecofriendly control of vectors mosquito is needed. In this regard botanical insecticide is looking more fruitful. Therefore, the present research aimed to investigate the effectiveness of methanolic extract and various fractions, including, n-hexane, ethyl-acetate, chloroform, and aqueous fraction, obtained from methanolic extract of Ailanthus altissima, Artemisia scoparia, and Justicia adhatoda using separating funnel against larval, pupal, and adult stages of Culex quinquefasciatus. The larvae and pupae of Culex quinquefasciatus were exposed to various concentrations (31.25-1000 ppm) of methanolic extract and its fractions for 24 hours of exposure period. For knock-down bioassay (filter paper impregnation bioassay) different concentration of the methanolic extract and its various fractions (i.e. 0.0625, 0.125, 0.25, 0.5 and 1mg/mL) were applied for 1 hour exposure period. The results were statistically analysed using standard deviation, probit analysis, and linear regression. The R2 values of larvae, pupae, and adult range from 0.4 to 0.99. The values of LC50 (concentration causing 50% mortality) for late 3rd instar larvae after 24 hours exposure period range from 93-1856.7 ppm, while LC90 values range from 424 -7635.5ppm. The values of LC50for pupae range form 1326.7-6818.4ppm and and values of LC90 range from 3667.3-17427.9ppm, respectively. The KDT50 range from 0.30 to 2.8% and KDT90 values range from1.2 to 110.8%, respectively. In conclusion, Justicia adhatoda may be effective for controlling populations of vector mosquito.


Por ser o vetor do vírus do Nilo Ocidental e da falaríase, o controle de Culex quinquefasciatus Say é provavelmente essencial. O tratamento com inseticida sintético parece ser mais eficaz para o controle dos mosquitos vetores. No entanto, esses produtos são tóxicos para o meio ambiente e organismos não visados. Consequentemente, o controle ecológico dos mosquitos vetores é necessário. Nesse sentido, o inseticida botânico parece mais produtivo. Portanto, a presente pesquisa teve como objetivo investigar a eficácia do extrato metanólico e de várias frações, incluindo n-hexano, acetato de etila, clorofórmio e fração aquosa, obtidos do extrato metanólico de Ailanthus altissima (Mill.) Swingle, Artemisia scoparia Waldst. & Kit. e Justicia adhatoda L. usando funil de separação contra os estágios larval, pupal e adulto de C. quinquefasciatus. As larvas e pupas de C. quinquefasciatus foram expostas a várias concentrações (31,25-1000 ppm) de extrato metanólico, e suas frações por 24 horas de período de exposição. Para o bioensaio knock-down (bioensaio de impregnação de papel de filtro), diferentes concentrações do extrato metanólico e suas várias frações (ou seja, 0,0625, 0,125, 0,25, 0,5 e 1 mg / mL) foram aplicadas por um período de exposição de 1 hora. Os resultados foram analisados estatisticamente usando desvio padrão, análise Probit e regressão linear. Os valores de R2 de larvas, pupas e adultos variaram de 0,4 a 0,99. Os valores de LC50 (concentração que causa 50% de mortalidade) para larvas de terceiro estádio tardio após 24 horas de período de exposição variaram de 93-1856,7 ppm, enquanto os valores de LC90 variaram de 424-7635,5ppm. Os valores de LC50 para pupas variaram de 1326,7-6818,4 ppm e os valores de LC90 variaram de 3667,3-17427,9 ppm, respectivamente. O KDT50 variou de 0,30 a 2,8% e os valores de KDT90 variaram de 1,2 a 110,8%, respectivamente. Por fim, a espécie J. adhatoda pôde ser eficaz para controlar populações de mosquitos vetores.

Resumen Introducción: Los manglares están en declive en el mundo, conocer su estructura y regeneración en diferentes niveles de alteración puede ayudar a comprender la dinámica poblacional ante situaciones de estrés. Objetivo: La meta principal del estudio fue analizar la estructura comunitaria y estimar la trayectoria de las poblaciones de Avicenia germinans en diferentes sitios de manglar que presentan tres niveles de alteración. Métodos: Ocho transectos de aproximadamente 500 m de largo y colocados de manera perpendicular a la laguna de Tampamachoco, Veracruz, interceptaron tres niveles de alteración en los sitios estudiados del bosque de manglar. En cada transecto, cuadrantes de 10 x 10 m, 5 x 5 m, y 1 x 1 m sirvieron para contar todos los árboles de mangle por especie y estadio de madurez en 2017 y 2018. Resultados: La especie más abundante fue: A. germinans seguida de Rhizophora mangle y Laguncularia racemosa. Con base en su abundancia, se encontraron diferencias significativas según los tres grados de alteración (AM R = 0.6, P < 0.001), ilustrado en un análisis de ordenación no métrico (nMDS). Una tabla de vida para A. germinans permitió un análisis poblacional que mostró una tasa intrínseca de incremento poblacional (r) negativa para los sitios que presentan alteración (-0.0027 u -0.0774) y positiva (0.0289) en el sitio sin alteración aparente. La supervivencia del estadio de plántula a juvenil varia de 50 % en el sitio no alterados a 5 y 4 % en los otros dos sitios. Conclusiones: Se necesitan medidas para incrementar la sobrevivencia de las plántulas y revertir la disminución de las trayectorias en los sitios con alteración, de otra forma el manglar tendrá una viabilidad reducida en el término medio.


Abstract Introduction: Globally, mangroves are declining; characterizing their structure and regeneration at different disturbance levels can help understand their responses to stressful situations. Objective: The study's primary goal was to analyze the mangrove community structure and to estimate the populations' trajectories of Avicenia germinans at different mangrove sites that present three levels of disturbance. Methods: Eight transects of approximately 500 m in length and laid perpendicular to the Tamapamchoco lagoon, Veracruz, intersected three levels of disturbance currently present on the mangrove forest. On each transect, 10 x 10 m, 5 x 5 m, and 1 x 1 m quadrants served to account for all mangrove trees by species and stage of maturity in 2017 and 2018. Results: The most abundant species was: A. germinans followed by Rhizophora mangle and Laguncularia racemosa. Based on their abundance, we found significant differences for the three levels of disturbance (ANOSIM R = 0.6, P < 0.001) presented in a non-metric ordination (nMDS). A life table for A. germinans allowed a population analysis that showed an intrinsic rate of population increase (r) that was negative at the disturbed sites (-0.0027 and -0.0774) and positive (0.0289) at the apparently undisturbed site. Seedling to juvenile stage survival ranged from 50 % at the undisturbed site to 5 and 4 % at the disturbed sites. Conclusions: Measures to increase the survival of seedlings are necessary to reverse the decreasing population trajectories at the disturbed sites; otherwise, the mangrove will have reduced viability in the medium term.

ABSTRACT

Introducción: el uso de las plantas medicinales es común en la población de Cuba. Pero, gran parte de la misma desconoce sus propiedades, forma de empleo, modo de aplicación, así como las contraindicaciones e interacciones con los medicamentos convencionales. En este contexto el adulto mayor juega un rol importante por los conocimientos empíricos y esotéricos adquiridos, además del empleo que hace de estas plantas. Objetivo: valorar el uso tradicional de plantas medicinales por el adulto mayor en la comunidad Lechuga de Arroyo Naranjo. Métodos: entrevista, basada en una guía de aspectos y temáticas específicas a adultos mayores, de la comunidad Lechuga de Arroyo Naranjo. Se obtuvo una lista de plantas medicinales con frecuente uso, sus aplicaciones, partes utilizadas, modos de empleo y conocimientos sobre sus contraindicaciones, toxicidad o interacciones con medicamentos convencionales. Resultados: se alegó el uso de 38 especies de plantas medicinales, agrupadas en 25 familias, el 71 % de las plantas informadas se cultivaban en los alrededores de las casas, el 65,9 % de las personas conoce un número importante de las propiedades y usos de las plantas medicinales, el 56,8 % expuso al menos una forma o tipo de preparación de las plantas identificadas. Se constató que las mujeres conocen más de las propiedades y usos de las plantas medicinales que los hombres, el 85,2 % no conoce las contraindicaciones, toxicidad, riesgos, así como las posibles interacciones con los medicamentos convencionales. Conclusiones: el adulto mayor en la comunidad de Lechuga, tiene conocimientos empíricos sobre el uso de plantas con propiedades medicinales, aunque es necesario la divulgación y educación de la población en relación con las propiedades útiles de las plantas, formas de preparación y contraindicaciones, toxicidad o interacciones con medicamentos convencionales.

ABSTRACT@#Methanolic extract from the leaves of Acanthus ilicifolius L. (A. ilicifolius L.) is a potent inhibitor of Candida albicans (C. albicans) growth and anti-inflammatory. C. albicans causes oral candidiasis in immunosuppressive condition. Mitogen-activated protein kinase (MAPK) signalling via p38 appears to discriminate between yeast and hyphal cells of C. albicans. Activation of p38 MAPK by hyphae results in the upregulation of proinflammatory cytokines. The p38 MAPK activation is known to impair corticosteroid action. The research was conducted to investigate the effect of methanolic extract A. ilicifolius L. treatment of oral candidiasis with the immunosuppressive condition through enhancement of p38 MAPK expression in the epithelial cells. Immunosuppressed conditions were obtained when 16 healthy male Rattus norvergicus (Wistar) was given oral administration of dexamethasone and tetracycline for 14 days and induced with C. albicans (ATCC-10231) 1 McFarland. The subjects were divided into four groups (n = 4/group): immunosuppression (IS), immunosuppression with oral candidiasis without treatment (ISC), immunosuppression with oral candidiasis and nystatin treatment (ISC+N), and immunosuppression with oral candidiasis and A. ilicifolius L. treatment (ISC+AI), and were treated for 14 days. Later, the rats were euthanised, and their tongue were biopsied. The p38 MAPK expression was subjected to immunohistochemical examination, observed under a microscope (400× magnification) and statistically analysed (one-way ANOVA, LSD-test, p < 0.05). The p38 MAPK expression of ISC+AI (36.05 ± 1.54) was higher than IS (26 ± 2.32), ISC (26.4 ± 3.71), IS+N (34.2 ± 0.99). Significant differences existed between ISC+AI and ISC+N to IS and ISC (p < 0.05). No significant differences were present between IS and ISC; ISC+AI and ISC+N (p > 0.05). Therefore, this treatment could enhance p38 MAPK expression in oral candidiasis with the immunosuppressed condition.

ABSTRACT Aim of the study To evaluate the role of micronized purified flavanoid fraction and ethanol Graptophyllum pictum extract in the treatment of anal ulcer. Method Twenty-eight Wistar rats were randomly allocated into four groups. Groups 2, 3 and 4 the anus were induced with croton oil, but was not induced on group 1. Groups 1 and 2 were treated with normal saline, while groups 3 and 4 were treated with micronized purified flavanoid fraction, and ethanol G. pictum extract, respectively. On 9th days blood sample were taken from the retro-orbital region, and Wistar was killed by cervical dislocation under ether anesthesia. The anal canal was resected up 2 cm from anal opening, weighted, photographically taken to measure the percentage of residual ulcer, and then prepared for microscopic examination. Elisa methods were done for superoxide dismutase and malondialdedhyde. The total leukocyte in the anal specimen was counted under 400 magnification power. superoxide dismutase, anal coefficient, and total leukocyte for statistical analysis were using ANOVA and LSD, while malondialdedhyde and percentage of ulcers were using Kruskal-Wallis and Mann-Whitney. Result Treatment with ethanol G. pictum extract dose of 100 mg/kg BW significantly reduces the percentage of anal ulcer, the edema, leukocyte infiltration, and malondialdedhyde, and increase the superoxide dismutase in comparison without treatment. Treatment with micronized purified flavanoid fraction did not reduce the leukocyte, anal coefficient, and percentage of anal ulcer, only increase malondialdedhyde and decrease superoxide dismutase significantly.


RESUMO Objetivo do estudo Avaliar o papel da Fração Flavonoica Purificada Micronizada e do Extrato Etanólico de Graptophyllum pictum no tratamento de úlcera anal. Método Vinte e oito ratos Wistar foram randomicamente alocados em quatro grupos. Nos grupos 2, 3 e 4, indução com óleo de cróton foi realizada no ânus, excetuando-se o Grupo 1. Os grupos 1 e 2 foram tratados com solução salina normal, enquanto os grupos 3 e 4 foram tratados com fração flavonoica purificada micronizada e extrato etanólico de Graptophyllum pictum, respectivamente. No nono dia, amostras de sangue foram colhidas da região retroorbital, e o rato Wistar sofreu eutanásia por deslocamento cervical sob anestesia com éter. O canal anal foi ressecado até 2 cm da abertura anal, ponderado e fotografado para medir a porcentagem de úlcera residual e, em seguida, preparado para exame microscópico. Os métodos superoxide dismutase e malondialdedhyde do ensaio Elisa foram realizados. A contagem total de leucócitos foi realizada na amostra anal com ampliação de 400 vezes. ANOVA e LSD foram utilizados para a análise estatística de superoxide dismutase, coeficiente anal e número total de leucócitos, enquanto os testes de Kruskal-Wallis e Mann-Whitney foram utilizados para a análise de malondialdedhyde e porcentagem de úlceras. Resultado O tratamento com o extrato etanólico de Graptophyllum pictum (100 mg/kg de peso corporal) reduz de modo significativo a porcentagem de úlceras anais, o edema, a infiltração de leucócitos e o malondialdedhyde e aumenta a superoxide dismutase, comparado ao não tratamento. O tratamento com a fração flavonoica purificada micronizada não reduziu os leucócitos, o coeficiente anal e a porcentagem de úlceras anais, apenas aumentou o malondialdedhyde e diminuiu significativamente a superoxide dismutase.

ABSTRACT

Diabetes is an independent risk factor for the development of cardiovascular disease, with approximately 80% of cardiovascular mortality and morbidity linked to vascular complications such as atherosclerosis. It has been estimated that up to one-third of patients with diabetes mellitus use some form of complementary and/or alternative medicine. One plant that has received attention from diabetic patients for its perceived antidiabetic properties is Clinacanthus nutans, a member of the Acanthaceae family that is known as snake grass. Ethnomedical applications of this herb have been identified for the treatment of certain conditions, including fever, diabetes, skin rashes, and insect bites. This review aims to assess the potential of C. nutans to be used in the prevention and/or treatment of diabetic vasculopathy. Evidence for antidiabetic, anti-inflammatory, and dyslipidemic properties of C. nutans, as shown from experimental studies, is presented and discussed. Diabetes, inflammation, and hyperlipidemia are known to play significant roles in the initiation and severity of diabetic cardiovascular disease; thus, targeting these factors might be beneficial for preventing and/or treating diabetic vasculopathy.

ABSTRACT

A new species of Alternaria causing leaf spots on the rubber tree (Hevea brasiliensis) in Yunnan, China, was isolated, examined, and illustrated. Morphologically, it belongs to the section Porri of Alternaria, which produces relatively large conidia and a simple or branched, filamentous long beak. It is, however, characterized by conidiophores gradually enlarging near the apex into a clavate conidiogenous cell and long ellipsoid to obclavate, smooth-walled conidia with a long filamentous beak. Molecular phylogenetic analyses based on ITS rDNA, GAPDH, and TEF1-alpha sequences demonstrate that the phytopathogen falls in the clade of the section Porri, being most closely related to A. sidae, A. sennae, A. deseriticola, A. cyamopsidis, A. rostellata, A. nitrimali, A. crassa, and A. thunbergiae.

ABSTRACT

Chorismate synthase(CS, EC:4.2.3.5) catalyses 5-enolpyruvy-shikimate-3-phosphate to form chorismate, which is the essential enzyme for chorismate biosynthesis in organisms. The amino acid sequences of CS from 79 species of higher plants were reported in GenBank at present. 125 amino acid sequences of CS from Baphicacanthus cusia and other 78 species of plants were predicted and analyzed by using various bioinformatics software, including the composition of amino acid sequences, signal peptide, leader peptide, hydrophobic/hydrophilic, transmembrane structure, coiled-coil domain, protein secondary structure, tertiary structure and functional domains. The phylogenetic tree of CS protein family was constructed and divided into eight groups by phylogenetic analysis. The homology comparison indicated that B. cusia shared a high homology with several plants such as Sesamum indicum, Nicotiana tabacum, Solanum tuberosum and so on. The open reading frame(ORF) of all samples is about 1 300 bp, the molecular weight is about 50 kDa, the isoelectric point(pI) is 5.0-8.0 which illustrated that CS protein is slightly basic. The ORF of CS we cloned in B. cusia is 1 326 bp, the amino acid residues are 442, the molecular weight is 47 kDa and pI is 8.11. The CS in B.cusia showed obvious hydrophobicity area and hydrophilicity area, no signal peptide, and may exists transmembrane structure areas. The main secondary structures of CS protein are random coil and Alpha helix, also contain three main structural domains which are an active structural domain, a PLN02754 conserved domain and a FMN binding site. The acquired information in this study would provide certain scientific basis for further study on structure-activity relationship and structure modification of CS in plants in the future.

ABSTRACT

This article explores the most recent evidence-based information on ethnomedicinal, phytochemical and pharmacological understanding of Hygrophila auriculata for the treatment of various diseases and health conditions. Various ethnomedicinal writings suggest the use of the plant or its parts for the treatment of jaundice, oedema, gastrointestinal ailments, diarrhoea, dysentery, urinogenital disorder, gall stones, urinary calculi, kidney stone, leucorrhoea, rheumatism, tuberculosis, anaemia, body pain, constipation, skin disease, and as an aphrodisiac. The plant has been reported to contain flavonoids (apigenin, luteolin, ellagic acid, gallic acid and quercetin), alkaloids (asteracanthine and asteracanthicine), triterpenes (lupeol, lupenone, hentricontane and betulin), sterols (stigmasterol and asterol), minerals, amino acids, fatty acids, aliphatic esters and essential oils. Extracts and bioactive compounds from the plant have been found to possess antimicrobial, anthelmintic, antitermite, nephroprotective, hepatoprotective, central nervous system protective, antitumour, antidiabetic, anticataract, antioxidant, haematopoietic, diuretic, antinociceptive, anti-inflammatory, antipyretic, antimotility, aphrodisiac, neuroprotection, anti-endotoxin and anti-urolithiatic activities. For this paper, we reviewed patents, clinical studies, analytical studies and marketed formulations from the earliest found examples from 1887 to the end of 2017.

ABSTRACT

The consumption of botanicals for therapeutic purposes has increased significantly in recent years. Drug-induced liver disease (DILI) is a frequent cause of acute liver injury, around 50% in the United States, and about 1% is secondary to the use of phytotherapeuticals and herbal supplies. Ruellia bahiensis, a plant species of the Acanthaceae family, is a tropical plant distributed in Northeastern Brazil. In folk medicine in the state of Bahia, the species is known as "mãe-boa" and is commonly used. L.S.S, a 23-year old, female, patient was admitted at University Hospital of Bahia-Brazil with signs and symptoms of acute hepatitis. She had made daily use of an herbal supply popularly known as "mãe-boa" for at least two years prescribed by a physician. Diagnostic investigation was negative for viral and autoimmune hepatitis, leptospirosis, dengue, and CMV (cytomegalovirus). The patient had to undergo liver transplantation. Explant revealed massive hepatic necrosis. According to histological findings, and after exclusion of other etiologies, liver damage was assigned to herbal supply. The prolonged use of Ruellia bahiensis infusions may have caused the liver dysfunction.

ABSTRACT

La diabetes es uno de los principales problemas de la salud pública mundial. En Venezuela se utilizan varias plantas para el tratamiento de la diabetes, sin embargo, los estudios farmacológicos de estas especies han sido insuficientes. Ruellia tuberosa L. (Acanthaceae) es una de estas plantas de uso en la medicina tradicional. En los últimos años, he mos estudiado la farmacología del extracto acuoso de la raíz de la R. tuberosa (RT) demostrando su actividad an ti - inflamatoria, antioxidante y protectora frente al daño renal inducido por la diabetes. El tratamiento subcrónico con este extracto fue capaz de inhibir el aumento de la glucosa sanguínea en ratas con diabetes inducida por la es - treptozotocina (ETZ). En este trabajo se evaluó el perfil fitoquímico preliminar y el contenido de polifenoles totales del extracto de RT, así como también sus efectos agudos sobre la glicemia en ratas diabéticas. Los resultados muestran que el RT produjo un efecto hipoglicemiante tanto en los animales controles como en las ratas con diabetes inducida por la ETZ, con porcentajes de variación de la glicemia comparables con el hipoglicemiante oral de referencia, la glibenclamida. Este extracto mostró la presencia de un alto contenido de polifenoles, lo que posiblemente se encuentre asociado con su actividad antidiabética y antioxidante, reflejada a través de la reacción del RT con el radical 2,2- difenil-1-picrilhidrazil (DPPH). Todos estos hallazgos contribuyen con la validación del RT como un extracto antidiabético, el cual involucran la disminución de la glicemia y el decremento del estrés oxidativo. Asimismo, sienta las bases para el aislamiento y caracterización de los componentes responsables de su actividad farmacológica.

ABSTRACT

Estudos que envolvem aspectos da polinização e sistema reprodutivo em Angiospermas são fundamentais para a compreensão de diversos processos ecológicos e evolutivos. Desta forma, foi realizada uma revisão bibliográfica com o objetivo de compilar informações sobre a polinização e o sistema reprodutivo encontrados na família Acanthaceae Juss. no Brasil. Um total de 30 estudos, com 31 espécies, fizeram parte da presente revisão. Quanto a distribuição geográfica e domínio fitogeográfico, a região Sudeste (50%) e o bioma Mata Atlântica (56,7%) apresentaram a maior proporção de estudos. Quatro síndromes de polinização foram registradas, sendo elas: troquilofilia, melitofilia, psicofilia e quiropterofilia, sendo a troquilofilia predominante (79,31% das espécies). Consequentemente, beija-flores foi o grupo mais comum de visitantes florais, interagindo com 86,2% das espécies, sendo Phaethornis pretrei o principal beija-flor visitante. Outros grupos de visitantes também foram registrados, como abelhas (segundo principal grupo), borboletas, morcegos, mariposas, vespas, moscas e besouros. Das 17 espécies estudadas no Brasil quanto ao sistema reprodutivo, todas apresentaram flores andróginas, sendo 16 espécies (94,1%) autocompatíveis. Fatores intrínsecos como a hercogamia e a dicogamia foram comuns em espécies de Acanthaceae, constituindo mecanismos que minimizam a interferência entre as funções masculina e feminina em flores andróginas e maximizam a polinização cruzada. Aproximadamente, 41,2% das espécies exibiram autopolinização espontânea, isto é, produção de frutos sem a necessidade de agentes polinizadores. Estratégias reprodutivas como a cleistogamia e a agamospermia também foram registradas nas espécies de Acanthaceae no Brasil. Contudo, mais estudos a respeito da polinização e sistema reprodutivo com acantáceas brasileiras precisam ser desenvolvidos, para ampliar a amostragem e, então subsidiar mais as estimativas desta revisão.


Studies involving aspects of pollination and breeding system in Angiosperms are fundamental in understanding many ecological and evolutionary processes. Thus, a literature review was conducted to aim of compiling information on the pollination and reproductive system in species of Acanthaceae Juss. in Brazil. A total of 30 studies, count 31 species, were part of this review. Regarding geographical distribution of studies, the Southeast Brazil (50%) and Atlantic Forest biome (56.7%) had the higher proportion of studies. Four pollination syndromes were recorded: trochilophily, melittophily, psycophily, and chiropterophily, in which trochilophily was predominant (79.31% of species). Consequently, hummingbirds were the most common flower visitors, interacting with 86.2% of the species, and Phaethornis pretrei was the main hummingbird visitor. Other groups of visitors were also recorded, such as bees (second main group, 34.5%), butterflies, bats, moths, wasps, flies and beetles. From the 17 species studied in Brazil about the reproductive system, all have androgynous flowers and 16 species (94.1%) selfcompatibility. Intrinsic factors such as herkogamy and dichogamy were common in species of Acanthaceae, constituting mechanisms that minimize interference between male and female functions in androgynous flowers and maximize cross-pollination. Approximately 41.2% of species presented spontaneous self-pollination, i.e. production of fruits without the need of pollinators. Reproductive strategies such as cleistogamy and agamospermy were also recorded in species of Acanthaceae in Brazil. However, more studies on the pollination and reproductive system with species of Acanthaceae in Brazil need to be developed to expand the sampling and then support better the estimates of this review.

ABSTRACT

La diabetes mellitus afecta a muchas personas alrededor del mundo, por lo que la comunidad científica está interesada en la búsqueda de nuevas drogas de origen sintético o natural para el tratamiento de la misma. En el presente trabajo se utilizaron cuatro plantas empleadas tradicionalmente en el tratamiento empírico de la diabetes y se realizó la síntesis total de los productos naturales α- yß- pentagaloilglucosa, ácido elágico y sus análogos. Las plantas estudiadas provienen de los géneros: Ouratea polyantha Engl. (Ochnaceae), Capraria biflora L. (Schrophulariaceae), Cassia fruticosa Mill. (Fabaceae) y Ruellia tuberosa L. (Acanthaceae). De la planta O. polyantha Engl., se aisló por HSCCC, e identificó por técnicas espectroscópicas el flavonoide rutina (OpC-4), la biflavona agathisflavona (OpD-5, OpB-M1), el ácido 4-hidroxibenzóico (OpD-10) y un nuevo megastigmano identificado como (6R,9S)-6'-(4''-hidroxibenzoil)-roseósido (OpD-9). Adicionalmente, se obtuvieron fracciones enriquecidas en lupeol y ß-sitosterol (OpBHex-FrCHCl3), agathisflavona (OpA, OpG) y rutina (OpA, OpC, OpG, OpH) las cuales fueron identificadas por CCF y HPLC comparando con muestras auténticas. Se evaluó el contenido de flavonoides totales como % de rutina en las fracciones OpC, OpG y OpH, por ensayos colorimétricos con AlCl3, obteniendo valores de: 7,08 ± 0,04; 31,9 ± 1,0; y 12,4 ± 0,3 %, respectivamente. De la planta C. fruticosa Mill., se aisló e identificó, por cromatografía flash y técnicas espectroscópicas el ester esteriodal ß-sitosterol palmitato•1/2•H2O (CfRS-3) y los flavonoides glicosilados kaempferol 3-O-rutinósido (nicotiflorin, CfH-1) y kaempferol 3-O-(2''-ramnosil)rutinósido (clitorin, CfH-2). Adicionalmente, se obtuvieron fracciones enriquecidas ß-sitosterol (CfB), y en los flavonoides nicotiflorin y clitorin (CfF, CfYY). De la planta C. biflora L., se aisló e identificó el glicosido manitol (CbE), el cual fue purificado por múltiples procesos de recristalización (CbM-2) y caracterizado por sus propiedades físicas y por la obtención de su derivado semisintético hexaacetilmanitol (CbM-2Ac). Adicionalmente, se obtuvieron fracciones enriquecidas en ß-sitosterol (CbI-FrCHCl3), azúcares (CbG, CbO), porfirinas tipo feoforbido-a (CbK y CbL) y terpenos aromáticos (CbK, CbF, CbN, CbP). De la planta R. tuberosa L., se obtuvieron fracciones enriquecidas en lupeol, betulina y ß-sitosterol (RtB, RtR-1, RtR-K), azúcares (RtI, RtR-G) polifenoles (RtJ, RtQac, RtSac, RtR-Mac, RtR-Nac) y terpenos aromáticos (RtQorg, RtSorg, RtR-Morg, RtR-Norg). En RtR-K se cuantificó por densitometría óptica el contenido porcentual de: lupeol 37,3 ± 0,8; ß-sitosterol 15,6 ± 0,6 y betulina (4,3 ± 0,3) % respectivamente. Los flavonoides aislados agathisflavona, nicotiflorin y clitorin mostraron inhibición significativa sobre el sistema enzimático G-6-Pasa microsomal cuyos valores fueron 63, 60 y 46 % respectivamente. Por otra parte, las fracciones terpenoidal (RtR-K) y porfirínicas (CbK y CbL) presentaron una inhibición moderada sobre la G-6-Pasa con 45, 25 y 27% respectivamente. Entre los productos sintetizados, los anómeros α- y ß-pentaacilglucosa (PAG, PBG y PGG-Bn) no presentaron actividad biológica significativa (<10%), lo cual indica claramente que el grupo galoilo es fundamental para la actividad biológica asociada a los taninos hidrolizables α-PGG y ß-PGG. A concentraciones finales de 50 µM, estos compuestos presentaron un porcentaje de inhibición de 77 y 79 % respectivamente con una significancia estadística importante (p<0,00006). Por otra parte, el ácido elágico (AE) sintetizado presentó un porcentaje de inhibición de 63% cuyo IC50 fue 76,50 µM, su derivado acetilado (AAcE) mostro un IC50 73,5 µM, mientras que, el derivado alquilico (AMeE) fue inactivo frente a la G-6-Pasa. Adicionalmente, AE y AAcE no mostraron actividad inhibitoria de la absorción intestinal de glucosa.

ABSTRACT

Justicia pectoralis Jacq., Acanthaceae, is a herb known popularly in Cuba as Tilo and used traditionally as sedative. The development in a solid pharmaceutical (Tablets 100 mg) using dry extract of Justicia pectolaris aqueous extract is of interest for the development of phytomedicines, which uses this active raw material. The aim of the present study was to carry out chemical and biological stability studies to the formulation. A method of coumarin determination by High Performance Liquid Chromatography (HPLC) was used and validated. The stability studies during different periods of time (24 months) showed a stability of the product stored at 32 ± 2 °C, and protected of the light.


Justicia pectoralis Jacq., Acanthaceae é uma erva conhecida popularmente em Cuba como Tilo e utilizada tradicionalmente como sedativo. O desenvolvimento de formas farmacêuticas sólidas (comprimido 100 mg) usando extrato aquoso seco de J. pectoralis é de interesse no desenvolvimento de fitoterápicos que empreguem esse princípio ativo. O objetivo do presente estudo foi realizar estudos de estabilidade químicos e biológicos da formulação. Um método de determinação de cumarinas por Cromatografia Líquida de Alta Eficiência (CLAE) foi usado e devidamente validado. Os estudos de estabilidade durante diferentes períodos de tempo (24 meses) mostraram a estabilidade do produto preservado a 32 ± 2 °C e protegido da luz.

ABSTRACT

This study was conducted to investigate the life history, morphology, and maturation of larval stages and adult worms of Fasciola gigantica in experimental mice. Lymnaea auricularia rubiginosa was used as the intermediate host, and Oryza sativa was used for encystment of the metacercariae, while Mus musculus was used as the definitive host for maturation study. Fresh eggs from the gall bladder of water buffaloes fully developed into embryonated ones and hatched out at days 11-12 after incubation at about 29masculineC. Free-swimming miracidia rapidly penetrated into the snail host, and gradually developed into the next larval stages; sporocyst, redia, and daughter redia with cercariae. Fully-developed cercariae were separated from the redia and shed from the snails on day 39 post-infection (PI). Free-swimming cercariae were immediately allowed to adhere to rice plants, and capsules were constructed to protect metacercariae on rice plants. Juvenile worms were detected in intestines of mice at days 3 and 6 PI, but they were found in the bile duct from day 9 PI. Juvenile and adult flukes were recovered from 16 mice experimentally infected with metacercariae, with the average recovery rate of 35.8%. Sexually mature adult flukes were recovered from day 42 PI. It could be confirmed that experimentally encysted metacercariae could infect and develop to maturity in the experimental host. The present study reports for the first time the complete life history of F. gigantica by an experimental study in Thailand. The obtained information can be used as a guide for prevention, elimination, and treatment of F. gigantica at environment and in other hosts.

ABSTRACT

The chemical structures of P1 A was identified by complete acid hydrolysis, partial acid hydrolysis, periodate oxidation-Smith degradation, methylation analysis, IR and NMR. The results showed that P1 A had a backbone consisting rhamnose, mannose, glucose and galactose. The side chain possessed arabinose and xylose. 1-->, 1-->6 and non-reducing terminal linkages existed in polysaccharide P1A, but there are doubling amount of 1-->2 and 1-->4 linkages. Oxidable linkage of P1 A accounted for 45%, and inoxidable linkage of P1A accounted for 55%. Mannose, glucose and galactose were mainly linked by 1-->2 linkage. Rhamnose, arabinose and xylose were mainly linked by 1-->2 and 1-->4 linkages. PlA contained beta-Glc(1,6)-,beta-Gal(1,3)-,beta-Man(1,4)-beta-Rha,-Glc(1,4)-, Glc(1)-,-Gal(1,4)- and Man(1)-.

AIM@#To study the gastroprotective effect and in vivo antioxidant potential of a standardized iridoid fraction from B. prionitis leaves (BPE) against different gastric ulcer models in rats.@*METHOD@#The standardized iridoid fraction from BPE at 50, 100, and 200 mg/kg body weight was administered orally, twice daily for 5 days for prevention from aspirin, ethanol, cold-restraint stress (CRS), and pylorus ligation (PL)-induced ulcers. Estimation of the antioxidant enzyme activity was carried out in a CRS-induced ulcer model, and various gastric secretion parameters including volume of gastric juice, acid output, and pH value were estimated in the PL-induced ulcer model.@*RESULTS@#BPE showed a dose-dependent ulcer protective effect in PL (18.67%-66.26% protection), aspirin (24.65%-63.25% protection), CRS (20.77%-59.42% protection), and EtOH (16.93%-77.04% protection)-induced ulcers. BPE treatment in PL-rats showed a decrease in acid-pepsin secretion, and enhanced mucin and mucosal glycoproteins. However, BPE reduced the ulcer index with significant decrease in LPO (P < 0.01-0.001), SOD (P < 0.01-0.001), and an increase in CAT (P < 0.01-0.001), activity in the CRS-induced model.@*CONCLUSION@#The data shows that the iridoid fraction from BPE possesses anti-ulcerogenic and antioxidant potential.

Aims: Barleria prionitis L. (Family Acanthaceae) is a medicinal plant found road side in India and whole plant or its various parts like leaves, root, bark, stem and flowers are used traditionally for various treatments like toothache, inflammation, boils, glandular swellings and ulcer. Leaf juice is useful in gastric ulcer. Here, we attempt to prove the use of this plant as gastroprotective agent. Study Design: This study was conducted to evaluate the antiulcer activity of methanol extract obtained from the leaves of Barleria prionitis Linn. Place and Duration of Study: The experiments were conducted at Pharmacology lab of Institute of Pharmaceutical Sciences, Kurukshetra University during the period of July 2012 to December 2012. Material and Methods: Antiulcer activity was performed using the protocols of ulcer induced by ethanol and indomethacin at two different doses (250 and 500mg/kg). Parameters like volume of gastric juice, pH, free acidity, total acidity, aspartate amino transferase (AST) and alanine amino transferase (ALT) were also determined in ethanol induced ulcer model. Results: The reduction in ulcer index in Barleria prionitis treated animals was found to be statistically significant (P=.05), when compared with control groups in both the models. Significant changes were observed in total acidity only at dose 500mg/kg only and changes were significant in AST, ALT levels at both the doses. Other parameters showed non-significant results. Conclusion: The results of the present study show that the methanolic extract of Barleria prionitis L. possess antiulcer activity. This work supports the traditional use of this plant in treating gastric ulcer.

ABSTRACT

<p><b>OBJECTIVE</b>To investigate the chemical compounds from the ethanol extract with inhibitory effects against aldose reductase from Thunbergia.</p><p><b>METHOD</b>Guided by anti-aldose reductase assay, compounds from the bioactive fraction (ethyl acetate extract) were separated and purified by various chromatographic methods including silica gel, Sephadex LH-20, and reversed-phase HPLC. Their structures were indentified based on analysis of the spectroscopic data including 1D and 2D NMR data.</p><p><b>RESULT</b>Eight compounds were obtained and identified as 8-hydroxy-8-methyl-9-methene-cyclopentane [7,11] -1,4, 6-trihydroxy-tetrahydronaphthalene-12-one, named as thunbergia A (1), 3,4-dihydro-4,5,8-trihydroxy-2-(3-methyl-2-butenyl) naphtha[2,3-b] oxiren-1(2H)-one (2), 8-(beta-gluco pyranosyloxy)-3,4-dihydro-2-(3-methyl-2-butenyl)naphtha [2,3-b] oxiren-1(2H)-one (3), galangin (4), quercetin (5), luteolin (6), 5,6,3',4'-tetrahydroxy -3,7-dimethoxy-flavone (7) and upeol (8).</p><p><b>CONCLUSION</b>Thunbergia A was a new derivative of tetrahydronaphthalene, and compounds 2 and 3 were separated from the genus Thunbergia for the first time.</p>
